Factors Affecting Callus Induction from Anther and Ovary of Okra (Abelmoschus esculentus L.)

Background: Okra [Abelmoschus esculentus (L.) Moench.] is a nutrient-rich vegetable crop widely grown in the tropics and sub-tropics mainly for its edible pods. The haploid technique has been used plant breeding improvements of plants to develop new varieties relatively short time. Hence, we have optimized several factors such as growth regulators (PGR), sucrose concentration, cold treatment, type media culture conditions callus induction from anther ovary okra (557 F1 hybrid). Methods: flower buds different sizes were collected determine various stages development then subjected pre-treatments. explants cultured on combinations PGRs i.e., naphthyloxy acetic (NOA), Indole acid (IAA), 2, 4-Dichlorophenoxy (2, 4-D), Benzyl amino purine (BAP), isopentenyl adenine 2iP, Kinetin (KIN) Thidiazuron (TDZ). Result: optimum developmental stage microspore initiation was achieved size about 11 mm long. Flower that emerged one week after flowering showed significantly higher percentage induction. ovule or two days prior anthesis 35±1mm. In conclusion, our study investigated effect affects future okra.
